Isolation and identification of bluetongue virus by first time in Cuba

Abstract

Bluetongue (BT) is a viral disease that affects domestic and wild ruminants. The etiological agent of this disease is bluetongue virus (BTV) which is mostly transmitted by blood-borne vectors of the Culicoides genus and has a global distribution. Cuba is located in a zone of endemic circulation of the virus. Although the presence of bovine reactors has been reported to the World Organization for Animal Health (WOAH) since 2007, there is no evidence of clinical manifestations of the disease and the virus has not been isolated. This work aimed to isolate and identify bluetongue virus from the blood of a naturally infected calf from Mayabeque province in 2018. The virus was isolated in chicken embryonated eggs and in cell culture from the blood of the calf at 21 days of age, which tested positive for the presence of virus by a nested reverse transcription polymerase chain reaction assay (nRT-PCR). The inoculated embryos showed embryonic mortality, decreased size, reddish coloration and/or edema as indicative of viral multiplication. In cell culture, rounded and refractile cell foci were observed. Virus identification was performed by nRT-PCR assays, a commercial real-time RT-PCR kit (rRT-PCR) and rRT-PCR based on SYBR Green-I for specific detection of BTV.
